以急性發(fā)熱性呼吸道癥狀為特征的退伍軍人癥是由吸入含有嗜肺軍團(tuán)菌的霧化水引起的。嗜肺軍團(tuán)菌血清組1zui常從患者中分離（1,2）。
IMMUNOCATCHTM軍團(tuán)菌（Legionella）是一種側(cè)流免疫層析法，可通過快速可靠地檢測尿樣中的抗原，對嗜肺軍團(tuán)菌血清型1感染進(jìn)行早期診斷。

原理

本產(chǎn)品是一種試劑盒，設(shè)計用于免疫層析法檢測尿樣中嗜肺軍團(tuán)菌血清型1 LPS抗原。
當(dāng)樣品通過毛細(xì)作用遷移通過結(jié)合墊時，樣品中的軍團(tuán)菌抗原與綴合至膠體顆粒的抗軍團(tuán)菌抗體結(jié)合形成抗原 - 抗體復(fù)合物?？乖?- 抗體復(fù)合物在試紙條上遷移，與固定在膜上的抗軍團(tuán)菌抗體特異性結(jié)合而產(chǎn)生紅線。然后通過肉眼觀察紅線的存在來確定軍團(tuán)菌抗原的存在。
此外，抗-Legionella抗體偶聯(lián)的膠體金在反應(yīng)中不被消耗，與固定在對照線上的抗免疫球蛋白抗體結(jié)合產(chǎn)生紅線，表明在測試條上的反應(yīng)是成功的。

【檢測方法】

1）從鋁袋中取出所需數(shù)量的磁帶，并將它們放在水平面上。

2）使用定量移液器慢慢地吸取90μL或更多的樣品。 通過一次移液操作，將全部樣品滴入盒子的樣品位置。 滴下后，剩余的標(biāo)本留在吸管內(nèi)。

3）在室溫（15-30°C）下將盒子放置15分鐘，

4）檢查盒子評估地點是否有線（標(biāo)有“C”和“T”）。

結(jié)果

應(yīng)該在反應(yīng)開始15分鐘后及時進(jìn)行評估。 檢查評估地點，并通過下面描述的紅線來評估結(jié)果。
1）正面：紅色控制線和測試線均在評估現(xiàn)場。 當(dāng)兩條線在反應(yīng)時間結(jié)束之前出現(xiàn)時結(jié)果是肯定的。

2、用做血清流行病學(xué)調(diào)查時，將標(biāo)準(zhǔn)抗原置中心孔，周圍1、3、5孔加標(biāo)準(zhǔn)陽性血清，2、4、6孔分別加待檢血清。待檢孔與陽性孔出現(xiàn)的沉淀帶*融合者判為陽性。待檢血清無沉淀帶或所出現(xiàn)的沉淀帶與陽性對照的沉淀帶*交叉者判為陰性。待檢孔雖未出現(xiàn)沉淀帶，但兩陽性孔的沉淀帶在接近待檢孔時，兩端均內(nèi)向有所彎曲者判弱陽性。若僅一端有所彎曲，另一端仍為直線者，判為可疑，需重檢。重檢時，可加大檢樣的量。檢樣孔無沉淀帶，但兩側(cè)陽性孔的沉淀帶在接近檢樣孔時變得模糊、消失，可能為待檢血清中抗體濃度過大，致使沉淀帶溶解，可將樣品稀釋后重檢。
3、用以檢測抗血清的效價時，將抗原置中心孔，抗血清倍比稀釋后置周圍孔，以出現(xiàn)沉淀帶的血清zui高稀釋倍數(shù)為該抗血清的瓊擴(kuò)效價。四、注意事項1、不規(guī)則的沉淀線可能是加樣過滿溢出、孔型不規(guī)則、邊緣開裂、孔底滲漏、孵育時沒放水平、擴(kuò)散時瓊脂變干燥、溫度過高蛋白質(zhì)變性或未加防腐劑導(dǎo)致細(xì)菌污染等所致。2、抗原抗體的比例與沉淀帶的位置、清晰度有關(guān)。如抗原過多，沉淀帶向抗體孔偏移和增厚，反之亦然?？捎貌煌♂尪鹊姆磻?yīng)液試驗后調(diào)節(jié)。由于產(chǎn)氣莢膜梭菌（Clostridium perfringens）廣泛存在于自然界中，且其毒素也可能存在于免疫過的或有自然抵抗力動物的腸道中。因此，在臨床上即使從腸道中檢測到產(chǎn)氣莢膜梭索菌的毒素，依然不能確定是產(chǎn)氣莢膜梭菌病。產(chǎn)氣莢膜梭菌病根據(jù)該病的流行病學(xué)特點、臨床及病理變化特征可做出初步診斷，確診則要進(jìn)行實驗室檢查。
